Has the Canon EOS C-mount adapter got a female thread? I wish to mount an EOS 5D on a trinocular Leica MZ APO microscope. Does this need a lens in the adapter?

The adaptor has a male thread on the C-Mount side and an EOS bayonet on the other side. Unfortunately we cannot answer your question about how to attach to your Leica microscope, if it needs a lens a.s.o. - we do not know this instrument

I want to attach my Celestron Neximage 10 to my Canon eos lenses. Canon eos cameras optical distance from the back of lens to the sensor is 44mm. The distance of this adapter is 26mm and the back focus of my Neximage is 13.1 mm. Would I need an extension tube 5 mm long to achieve focus? Thank you, David.

Yes - even if you might be able to reach focus without the 5mm extension, the sensor would be so far away from the distance for which the lens is designed, that the quality would suffer.
So, we highly recommend the extension.
